摘要
延胡索乙素是中药元胡中镇痛作用较强的有效成分,为了解药动学与药效学的关系,本文采用灵敏度高、选择性强的胶束荧光法,研究了延胡索乙素在兔血浆中的药代动力学。实验结果表明,延胡索乙素在兔体内的代谢属于吸收快、消除慢、能保持较长时间内有镇痛作用的药物,其几个主要药动学参数为t_(1/2)(α)为1.98h、t_(1/2)(β)为16.7h、t_(1/2)(ka)为0.702h、K_(21)为0.238h ̄(-1)、K_(10)为0.061h ̄(-1)、K_(12)为0.093h ̄(-1)、AUC为34.66μg·h/ml。
Tetrahydropalmatine is an effective component possessing analgesic effect in Cordalis yanhusuo. Its pharmacokinetics in rabbit was studied by using highly sensitive and selective micellar fluorometry. The results suggested that the metabolism of tetrahydropalmatine was characterized by rapid absorption slow elimination and long lasting analgesic effect.The main pharmacokinetic parameters are t_(1/2)(α)=1. 98h,t_(1/2)(β)=16.7h,t_(1/2)(Ka)=0.702h,K_(21)=0.238h ̄(-1),K_(10)=0.061h ̄(-1),K_(12)=0.093h ̄(-1) AUC 34.66μg/ml·h.
