摘要
本文观察了小鼠肺腺癌LA795母系及不同转移潜能克隆株LA1、LA5细胞其胞内[Ca ̄(2+)]及诱导分化剂维胺酸和二甲基亚砜对其影响。发现母系与克隆株细胞间,其胞内静息[Ca ̄(2+)]确有差异(P<0.01)。中转移LA5克隆株胞内[Ca ̄(2+)]最高,不转移LA1克隆株最低,母系LA705胞内[Ca ̄(2+)]居中。在克隆株细胞中,胞内[Ca ̄(2+)]与其转移潜能成正比。维胺酸作用不同时间,LA795、LA1、LA5胞内[Ca ̄(2+)]先下降,然后逐渐恢复用药前水平,最终至胞内[Ca ̄(2+)]升高。二甲基亚砜作用后,引起胞内[Ca ̄(2+)]一过性升高,然后下降最后又升高。二者均终至所用细胞内[Ca ̄(2+)]升高,提示其诱导分化作用过程中,[Ca ̄(2+)]变化起作用,胞内[Ca ̄(2+)]与肿瘤转移性有一定关系。
The intracellular [Ca ̄(2+)] of mouse pulmonary adenocarcinoma LA795cell line and its clone LA1,LA5 were measured using Fura-/AM as fluorescent indica-tor.The results indicated that [Ca ̄(2+)]_i was varied.For the clones(nonmetastaticLA1,medium metastatic LA5) [Ca ̄(2+)]_i was positively correlated with their metastaticpotential.RⅡ(4-hydroxycarbonphenyl retinoamide)is an differentiation inducer.10 ̄(-5)MR Ⅱ temporarily decreased [Ca ̄(2+)]_i (6h)of all tested cells,then (3d) [Ca ̄(2+)]_i re-bound,and finally (7d)[Ca ̄(2+)]_i increased,DMSO (dimethyl sulfoxide)induced[Ca ̄(2+)]_i to ascend (6h)in all cells,then(3d)[Ca ̄(2+)]_i descended, andfinally(7d)[Ca ̄(2+)]_i decreased for LA795 cell,increased for LA1 and LA5 cells.The results suggested that [Ca ̄(2+)] play a role in the function of retinoamide.
