摘要
本文报道了在8淋巴细胞杂交瘤技术中采用脾内微量免疫及腹腔植入联合免疫方法获得了两株分泌抗神经肽Y(NPY)单克隆抗体(McAb)的杂交瘤细胞系,用ELISA法测定腹水McAb的效价为10 ̄(-3)~10 ̄(-5)。ELISA交叉试验结果表明,该McAb不与ACTH反应,两种McAb的亲和常数(Ka)各为1×10 ̄9M ̄(-1)和5×10 ̄7mol/L ̄(-1),抗原决定簇分析结果表明此两种McAb针对不同抗原决定簇。
his article reported a new immunization protocol for
production of McAb against 36 aminoacids NPY. Firstly intrasplenic
innoculation of affinity-beads of NPY-sepharose 2B was per-formed on
BL ab/c mice,then the NPY-binded nitrocellulose filter disks were
twicely implantedintraperitoneally,3 days prior to fusion the
intrasplenic injection was repeated once again.Afterfusion、selection
and cloning procedures, 2 hybridoma cell lines were successfully
obtained.TheMcAb titer from ascitic fluid were about 10 ̄3 ~ 10 ̄5
by ELISA test. Their affinity constantwere 1× 10 ̄9mol /L(-1)and 5×
10 ̄7mol/L ̄(-1) separately. They were very specific to NPY, but
notcross-react with unrelated peptides. After analysis of epitope
specificity, it is evident that theMcAb was directed at different
epitopes.
