羊开口提取物的分离及对艾滋病毒逆转录酶和DNA多聚酶活性的抑制作用 被引量:5

Differential Inhibitory Effects and Separation of Melastoma normale Extracts on the Activities of HIV Reverse Transcriptase and DNA Polymerase

摘要 羊开口经提取分离为Y-1、Y-P、Y-A、Y-E、Y-A-1和Y-A-1和Y3~11等部分,通过其对艾滋病毒逆转录酶(HIV-RT)和DNA多聚酶(DNAPol)活性试验,发现Y-1、Y-A、Y-A-1、Y-9和Y-11有极强的抑制酶活性作用。50%酶活性抑制浓度(IC50)对HIV-RT分别为0.1~0.5μg/ml,对DNApolα为0.5~2.5μg/ml。HIV-RT的抑制率当浓度Y-A、Y-A-12.5μg/ml,Y-9、Y-112μg/ml和Y-15μg/ml时分别为99.37%,93.35%,98.24%,99.92%和90.0%:DNApolα的抑制率当Y-A、Y-9、Y-11的浓度为2.5μg/ml和Y-1浓度为5μg/ml时,分别为95.45%,93.33%,93.01%和99.11%。结果提示羊开口中存在抑制HIV-RT和DNApolα的物质。 The extracts of Y-1、Y-P、Y-A、Y-E、Y-A-1 and Y-3-11 from Melastoma normale were tested for ability to inhibit the activies of HIV reverse transcriptase and human deoxyribonucleic acid(DNA) polymerase.Among 15 fractions,Y-1、Y-A、Y-A-1、Y-9 and Y-11 showed very strong inhibition enzyme activities.The 50% inhibition concentrations(IC50) 0.1~0.5μg/ml for HIV reversetranscriptase and 0.5~2.5μg/ml for DNA polymerase α.HIV reverse transcriptase was inhibited by 99.37%,93.35%,98.24%,99.92%and 89.68% in the presence of Y-A、Y-A-1(2.5μg/ml),Y-9、Y-11(2 μg/ml)and Y-1(5 μg/ml) respectively.DNA polymerase αwas inhibited by 95.45%,93. 33%, 93.01% and 99.11% in the presence of Y-A、Y-9、Y-11(2.5 μg/ml)and Y-1(5μg/ml)respectively.These results indicated that the M.normale contained as vet unidentified substance which inhibited the activities of HIV reverse transcriptase and cellular DNA polymerases.
