微小RNA-7对人肺癌95D细胞体外增殖的作用 被引量:19

The effects of miRNA-7 on the proliferation of human lung cancer cell line 95D cells

摘要 目的:探讨微小RNA-7(microRNA-7,miR-7)对人肺癌95D细胞体外增殖的作用。方法:采用体外转染法将miR-7瞬时转染95D细胞后,应用Real-time PCR特异探针法检测95D细胞中miR-7的表达情况,应用MTT法检测95D细胞的生长情况,FCM法分析细胞周期变化,并用Western印迹法检测95D细胞中表皮生长因子受体(epidermal growth factor receptor,EGFR)表达的变化。结果:与对照组相比,miR-7转染组95D细胞的miR-7表达水平显著增加(P<0.05),细胞生长明显受抑(P<0.05),且主要阻滞在G1期(P<0.05),而EGFR表达显著下调。结论:miR-7可以抑制人肺癌95D细胞的体外增殖,可能作为后续肺癌生物治疗的新靶点。 Objective:To explore the effects of miRNA-7(miR-7) on the proliferation of human lung cancer cell line 95D in vitro.Methods:MiR-7 was transiently transferred into 95D cells using FuGENE-HD reagent in vitro.The expression level of miR-7 in 95D cells was determined using specific probe of real-time PCR before or after transfection.The proliferation of 95D cells was accessed using MTT assay.The cell cycle was analyzed by flow cytometry.The expression of epidermal growth factor receptor(EGFR) was determined by Western blot.Results:The expression level of miR-7 was significantly increased in miR-7-transfected 95D cells compared with that in control group(P<0.05).The proliferation of miR-7-transfected 95D cells was significantly inhibited.The cells were arrested at G1 phase(P<0.05).The expression of EGFR was markedly down-regulated(P<0.05).Conclusion:MiRNA-7 inhibited the proliferation of 95D cells,suggesting that miRNA-7 might be a novel target for the biological therapy of human lung cancer.
