邪毒壅盛证H_(22)荷瘤小鼠胸腺独特上调的高表达基因

Specially Up-Regulated and Highly Expressed Genes in Thymus of the H_(22) Tumor-Bearing Mice with Pathogenic Toxin-Exuberance Syndrome

摘要 目的观察邪毒壅盛证荷瘤小鼠胸腺基因表达的特征。方法综合采用荷瘤小鼠及其标准化辨证方法,采用Gene-Chip Mouse Exon 1.0 STArray检测不同证候H22荷瘤小鼠胸腺组织基因的表达,并分别筛选2006年及2009年两批数据中邪毒壅盛证独特高表达的基因,进行理论研究。结果 2006年、2009年早期邪毒壅盛证荷瘤小鼠胸腺组织一致独特上调的基因108个,取各自最高表达的15个基因,有8个重叠:Trim30、Sept6、Mcm3、Supt16h、Vps35、Smc3、Rsl1d1、Pa2g4,另分别有Smc1a、Eif3s10、Phf6、Sesn1、Iws1、Lin7c、Itch,以及Tax1bp1、Ifi204、Pnn、Ptbp2、Ascc3、Ddx10、Luc7l2等非重叠基因,这些基因主要涉及细胞的增殖、基因的表达与调控。结论邪毒壅盛证荷瘤小鼠胸腺高表达的基因多涉及促进基因的表达与复制,这与该证候肿瘤增长迅速相关,提示胸腺尚处于被动且积极的免疫代偿状态;鉴于与邪毒壅盛证同期的气虚证,以及与其它对照比较,这些基因独特上调,提示荷瘤小鼠同病异证在胸腺层面是有其物质基础的。 Objective To investigate the characteristics of the gene expression in thymus of the H22 tumor-bearing mice with pathogenic toxin-exuberance syndrome.Methods By the quantitative four diagnosis and syndrome differentiation methods and Gene Chip Mouse Exon 1.0ST Array,we detected the thymus Gene Expression of H22 Tumor-bearing Mice with different syndromes,and screened out the up-regulated genes of pathogenic toxinexuberance syndrome in the year of 2006'and 2009'.Results In the year of 2006 and 2009 data there were 108 up-regulated genes in uniform of pathogenic toxin-exuberance syndrome which happened on the tumor early stage.Among the expression of the fifteen highest genes,there were eight genes which were the same in the two different years'experiment.They were trim30,S-ept6,Mcm3,Supt16h,Vps35,Smc3,Rslld1 and Pa2g4.The other genes,such as Smcla,Eif3s10,Phf6,Sesn1,Iws1,Lin7c,ltch,TAX1BP1,Ifi204,Pnn,Ptbp2,Ascc3,Ddx10 and Luc712 were different.These genes were most about cell proliferation,the expression and regulations of genes.Conclusion ①The highly expressed genes in theymus of pathogenic toxin-exuberanec syndrome are most about the expression and replicatio of genes.These are connected with the fast growth of tumor of this syndrome.It also means that the thymus are in a state of passive and active immunity comprensation.②These genes are specially expressed in pathogenic toxin-exuberance syndrome compared with the pideficiency syndrome which is also happened on early stage,and with other syndromes,it shows that different syndromes of H22 tumor-bearing mice have very different gene expression in thymus.
