鸽热休克蛋白70基因的扩增与同源性比较

摘要 [目的]扩增鸽热休克蛋白(HSP)70基因,并与其他禽类作同源性比较。[方法]参考禽类的HSP70基因序列设计、合成引物,从热应激鸽肝脏组织中提取总RNA作为模板,RT-PCR扩增目的基因。[结果]RT-PCR扩增到的基因片段为576 bp,经测序,确定为鸽HSP70基因;鸽HSP70基因与鸡、鹌鹑、珍珠鸡同类基因序列的同源性达97%以上。[结论]禽类的HSP70基因具有较高保守性。 [Objective] To amplify and do homology comparison of heat shock protein(HSP)70 gene in pigeon.[Method] A pair of primers were designed and synthesized using HSP70 gene sequence in poultry as the reference sequence.Total RNA was extracted from livers of heat-stressed pigeons,and the objective gene was amplified by RT-PCR.[Result] The gene fragments amplified by RT-PCR was about 576 bp,and was confirmed as HSP70 gene in pigeon by sequence measurement.The homology of HSP70 gene in pigeon was over 97%,compared ...
