摘要
目的从植物浸膏中筛选对乙酰胆碱酯酶有抑制作用的物质,为研发治疗阿尔茨海默病(AD)的新药提供先导化合物。方法建立乙酰胆碱酯酶抑制剂(AchEI)体外筛选模型,用改良Ellman比色法测定AchE的活力,随机筛选多种植物浸膏,对有抑制作用的浸膏进一步进行活性追踪,筛出活性物质。结果追踪得抑制率超50%的两个单体化合物(东茛菪内酯和cannabichromeorcinic acid)。结论东茛菪内酯和cannabichromeorcinic acid可作为治疗AD的先导化合物进行深入研究。
OBJECTIVE To screen out AchE inhibitors from herb extracts. The inhibitors will be used as lead compounds to develop new drugs for Alzheimer’s disease( AD) . METHODS An AchEIs screening model was established in vitro. The extracts of herbs were randomly screened with the improved Ellman colorimetry. The inhibitors were traced from active extracts.
