摘要
本文从PCR技术入手,以人基因组DNA为模板扩增出人神经营养素-3(humanneurotrophin-3,hNT-3)全长基因,并对克隆至pUC18质粒中的hNT—3基因做了全序列分析。序列分析结果表明,除hNT—3中第130位编码prepro-序列-95位Leu密码子的第一个碱基外(C→T),其余序列同文献报道完全一致,改变的碱基并不影响其编码的氨基酸序列。
Human Neurotrophin-3 intact gene was obtained by polymerase chain reaction using human genomic DNA as a template. The fragment was cloned into pUC18 and then sequenced. DNA sequencing results showed that except one base locating at the first place of the code encoding the-95 Leu in the prepro-sequence (position 130, C to T), the gene sequence was identical to that had been reported before,and the changed base had no influence on the amino acid which it encoded.
