LIG4mRNA与XRCC4mRNA在宫颈癌中的表达及临床意义 被引量:1

摘要 目的探讨宫颈癌组织中DNA连接酶4(LIG4)及X射线修复交叉互补基因4(XRCC4)的mRNA表达水平及临床意义。方法应用实时荧光定量RT-PCR技术检测61例宫颈癌和20例正常宫颈组织中LIG4和XRCC4的mRNA表达水平,分析两种基因的表达与宫颈癌分期、分化程度及盆腔淋巴结转移之间的关系。结果 LIG4 mRNA和XRCC4mRNA在宫颈癌组织及正常宫颈组织中的表达分别为(2.88±1.97)、(5.02±1.87)和(2.16±0.99)、(3.72±1.36),差异有统计学意义,P值分别为0.00和0.00;宫颈癌患者盆腔淋巴结有转移组和无转移组中的表达分别为(0.61±0.17)、(0.41±0.28)和(0.80±0.31)、(0.58±0.26),差异有统计学意义,P值分别为0.00和0.01;宫颈癌低分化组和中高分化组中的表达分别为(1.81±0.99)和(2.03±1.25)、(1.50±0.76)和(1.45±0.57),差异无统计学意义(P>0.05)。LIG4 mRNA在Ⅱ期和Ⅲ期宫颈癌组织中的表达分别为(0.71±0.34)和(0.96±0.36),差异有统计学意义,P=0.01。LIG4 mRNA与XRCC4mRNA在宫颈癌组织中表达呈正相关(r=0.38,P=0.00)。结论 LIG4 mRNA和XRCC4 mRNA在宫颈癌组织中的异常表达与宫颈癌的发展及盆腔淋巴结转移密切相关。 Objective To explore the expression levels and the clinical significance of DNA 1igase IV(LIG4) mRNA and X-ray repair cross complementing gene 4(XRCC4) mRNA in cervical carcinoma tissues.Methods The expression of LIG4 mRNA and XRCC4 mRNA were detected by real-time quantitative reverse transcription-polymerase chain reaction(QRT-PCR) method in 61 cervical carcinoma tissues and 20 normal cervical tissues.The relationship were analysed between the expressions of the two genes and stage,histodifferentiation and pelvic lymph node metastasis of cervical carcinoma.Results The expression of LIG4 mRNA and XRCC4 mRNA in cervical carcinoma tissues and normal cervical tissues were respectively(2.88 ± 1.97) and(5.02 ± 1.87),(2.16 ± 0.99) and(3.72 ± 1.36).The differences showed statistical significance(P = 0.00 and P = 0.00,respectively).The expression of LIG4 mRNA and XRCC4 mRNA in cervical carcinoma tissues of pelvic lymph node metastasis and no pelvic lymph node metastasis were respectively(0.61 ± 0.17) and(0.41 ± 0.28),(0.80 ± 0.31) and(0.58 ± 0.26).The differences showed statistical significance(P = 0.00 and P = 0.01,respectively).Their expression in cervical carcinoma tissues of low-histodifferentiation and high-histodifferentiation were respectively(1.81 ± 0.99) and(2.03 ± 1.25),(1.50 ± 0.76) and(1.45 ±0.57).The differences have no statistical significance(P >0.05).The expression of LIG4 mRNA in cervical carcinoma tissues of stage Ⅱ and stageⅢ was respectively(0.71 ± 0.34) and(0.96 ± 0.36).The difference had statistical significance(P = 0.01).There was not correlation between expression of LIG4 mRNA and XRCC4 mRNA in cervical carcinoma tissues(r = 0.38,P = 0.00).Conclusion Abnormal expression of LIG4 mRNA and XRCC4 mRNA in cervical carcinoma tissues was relevant to the progression and pelvic lymph node metastasis of cervical carcinoma.
