摘要
目的:建立知母中菝葜皂苷元的含量测定方法,为其提供质量标准。方法:应用高效液相色谱-蒸发光散射检测器,Di-amonsil C_(18)(150mm×4.6mm,5μn)色谱柱,以甲醇-水(95:5)为流动相,流速为0.8 mL·min^(-1),漂移管温度为90℃,载气流速为1.5L·min^(-1)。结果:菝葜皂苷元在1.04~5.22μg范围内呈良好线性关系,平均回收率为97.6%(RSD=2.1%)。分析了湖北、山西及辽宁等产地知母中菝葜皂苷元的含量,不同产地知母中菝葜皂苷元的含量为0.91%~2.94%。结论:该方法简便、准确,无干扰,可用于知母中菝葜皂苷元的的质量控制。
Objective : To establish the quantitative method for sarsasapogenin in Anemarrhena asphodeloides Bge. by HPLC -ELSD. Method:The column used was a packed with 5 μm stationary phase Diamonsil C18 (4.6 mm ×150 mm,5μm). The mobile phase consisted of methanol- water(95:5),with a flow rate of 0. 8 mL · min^-1 ,the contents of sarsasapogenin were measured. At a temperature of drift tube of 90℃ and the nebulizer nitrogen flow rate was 1.5 L · min^-1. Result: The linear range of the component was 1.04 - 5.22μg. Several samples of Anemarrhena asphodeloides Bge. were analyzed ; The contents in crude form from different areas were 0.91% - 2.94%. The average recovery of sarsasapogenin in crude material was 97.6% ( RSD = 2. 1% ). Conclusion: The method is reliable, simple and precise ,and it can be used for the quality control of Anemarrhena asphodeloides Bge.
