摘要
目的 研究新型β受体阻滞剂卡维地洛(Car)对单个核细胞表达CD25的影响。方法 分离外周血单个核细胞(PBMC),以PMA+A23187作为刺激剂,分组体外培养。用MTT方法检测PBMC的活性;用流式细胞仪检测单个核细胞的凋亡率和CD25的表达水平。结果 (1) 20μmol以内的Car对PBMC没有毒性; (2)Car能显著抑制PBMC表达CD25, 24h时,浓度3组为(8. 23±267)%,与对照组(30. 73±4. 10)%比较,差异有显著性(P<0. 01)。结论 Car可以抑制PBMC表达CD25。这可能是卡维地洛治疗慢性心衰的机制之一。
Aim To investigate the effect of carvedilol,a new-type beta-blocker,on the expression of CD25 peripheral blood mononuelear cells in vitro. Methods Peripheral blood mononuclear cells(PBMCs) were isolated from healthy volunteers ,and incubated with phorbol 12-myristole 13-acctate(PMA) and A23187 with or without carvedilol in vitro. After incubation,The viability of PBMCs was detected by MTT;The expression of PBMC activation markers such as CD25 on cell surface was assayed with flow cytometry. At the same time , the early and late apoptosis of PBMC was assayed by flow eytometry. Results ( 1 )There was no significant toxicities on examined human peripheral blood mononuclear cells when carvedilol was used at concentrations ≤20 μmol; (2)Car could significantly inhibit the expression of CD25 on PBMC surface. After 24 hours,the expression of CD25 in Car group was (8.23 ±2.67)% ,while that of the control group was (30.73 ±4.10)%. Conclusions The expression of CD25 on PBMC was inhibited by car. It may be to explain in part its therapeutic mechanisms in chronic heart failure.
