摘要
目的采用酶法提取杜仲中降压活性成分.方法选用纤维素酶,通过L9(3 4)正交试验确定京尼平苷酸和京尼平苷最佳提取工艺条件.结果提取京尼平苷酸的最佳工艺参数:温度45℃,介质pH=5,酶解时间1 h,每100 g杜仲需纤维素酶0.3 g,提取率达到1.58%;提取京尼平苷的最佳工艺参数为:温度45℃,介质pH=7,酶解时间0.5 h,每100 g杜仲需纤维素酶0.2 g,提取率达到0.26%.结论与传统水浸提取工艺相比,提取率提高9.09%~14.45%.
Purpose The extraction of hypotensive constituents from E. ulmoides was studied by enzymolysis. Methods Cellulase was chosen to carry out the experiment and the optimum extraction conditions were determined by orthogonal design method L9(3^4). Results The optimum extraction condition of GPA was as follows: 45℃ as extraction temperature, 5.0 as pH of solvent, 1 h as extraction time and 100:0.3 as optimum solid-enzyme ratio. While the optium extraction condition of GPS was as follows: 45 ℃ as extraction temperature, 7.0 as pH of solvent, 0.5 h as extraction time and 100:0.2 as optimum solid-enzyme ratio. Under these conditions, the extraction percentage of GPA and GPS was 1.58% and 0.26% respectively. Conclusion The extraction percentage of GPA and GPS using above method was higher than that of water extraction.
