摘要
目的观察表没食子儿茶素没食子酸酯(EGCG)对糖尿病大鼠肾脏损害的治疗作用及糖尿病大鼠肾氧化应激的影响。方法采用链脲佐菌素(STZ)诱导大鼠糖尿病模型,观察12周时肾脏损害和氧化应激改变,以及给予EGCG(2.5mg/kg和5mg/kg)治疗后的变化。结果12周时糖尿病大鼠与正常对照组比较:肾质量指数显著增加,肾脏的抗氧化能力显著降低且氧化应激增强;EGCG治疗组与糖尿病组比较:肾指数显著降低,总抗氧化能力和肾脏超氧化物岐化酶活性显著提高,脂质过氧化产物丙二醛(MDA)显著降低。结论糖尿病大鼠与正常大鼠相比抗氧化能力降低且氧化应激增强,EGCG可显著提高糖尿病大鼠肾脏的抗氧化能力和降低氧化应激,对糖尿病大鼠肾脏具有保护作用。
Objective To investigate the influence of EGCG(epigallocatechin-3-gallate) on renal malondialdehyde ( MDA), SOD, GSH-Px in the rat with type Ⅱ diabetics. Methods SD rats were divided into the con- trol group,STZ-treated group,EGCG-treated group Ⅰ and EGCG treated group Ⅱ. First, the rats in the latter three group were induced to be diabetic nephropathy by intraperitoneal injection with 65mg/kg streptozoticin (STZ) ,and then,EGCE was continuously given 2.5mg · kg^-1· d^-1 and 5mg · kg^-1·d^-1 4 weeks after STZ injection in EGCG-treated group respectively. The MDA, SOD, GSH-Px in the rat were observed respectively. Resuits At 12 weeks diabetic rats vs control group:the renal index of kidney increased significantly, the antioxidation capability of kidney decreased significantly. No significant difference of blood glucose was found between diabetic rats EGCG treated, gruops. Comparing with diabetic group, the levels of blood lipid, serum creatinine, urinary albumin excretory rate ,renal MDA ,plansm Hcy in EGCG treatd groups decreased significantly,but the renal SOD and GSH-Px increased markedly. Conclusion EGCG is effectively in improving the biahemical parameters and renal pathologic change. EGCG possesses antioxidant properities that can increase the antioxidant capability of kidney, decrease oxidative stress.
