摘要
目的探讨钙调神经磷酸酶(C aN)在肾血管性高血压心肌肥厚发展中的作用,以及C aN抑制剂环孢菌素A(C sA)对心肌肥厚的影响。方法39只大鼠(造模组)采用两肾一夹法行肾血管性高血压大鼠心肌肥厚造模,2个月时处死15只(造模Ⅰ组);余24只中,13只予生理盐水腹腔注射(造模Ⅱ组),11只行C sA腹腔注射(造模Ⅲ组)1个月后处死。另设22只大鼠为对照组,除不放置银夹外,其他处理与造模组相同,分别于2个月(对照Ⅰ组)、3个月(对照Ⅱ组)各处死11只。观察各组大鼠心肌肥厚程度、C aN mRNA和蛋白质表达及C aN活性的改变。结果左室重量(LVW)及与胫骨长度的比值(LVW/TL),光镜下心肌细胞横截面积(A rea),心肌C aNmRNA和蛋白质表达及其活性在造模Ⅰ组、Ⅱ组均较对照Ⅰ组、Ⅱ组增高(P均<0.05),造模Ⅲ组较造模Ⅰ组、Ⅱ组均显著下降(P均<0.05)。结论C aN参与肾血管性高血压大鼠心肌肥厚发展;抑制C aN活性可逆转心肌肥厚。
Objective: To investigate the activity of calcineurin in the development of cardiomegaly with renovasular hypertension, and to determine the effect of calcineurin inhibitorcyclosporine A (CsA)on the cardiomegaly. Methods:39 rats of renovascular hypertension which induced by two kidney-one clip mehtods were divided into 3 groups. M Ⅰ group (n= 15) were sacrificed after 2 months,M Ⅱ group (n= 13) received peritoneal injection with 0. 9% Nacl per day,then sacrificed after 1 month;MⅢ group (n= 11) received peritoneal injection with CsA per day,then sacrificed after 1 month. Control group (n = 22) were performed with the same method except the silvery clip,then divided into 2 groups equally(C Ⅰ and C Ⅱ )and sacrificed after 2 and 3 months. Results:The ratio of the left ventricular weight to tibial length (LVW/TL),the area of cardiac myocyte,mRNA and protein expression and the activity of calcineurin were significantly higher in M Ⅰ and M Ⅱgroups than that in CⅠ and CⅡi groups, which of MⅢ group were significantly higher than that in M Ⅰ and M Ⅱ groups. Conclusions :Calcineurin plays a role in the progression of cardiac hypertrophy in renovascular hypertensive rats. The inhibition of calcineurin can reverse cardiac hypertrophy.
