肝纤维化大鼠肝组织TGF-β_1表达和细胞外胶原沉积及中药抗肝纤维化治疗影响 被引量:2

摘要 目的探索中药丹芍化纤胶囊治疗CCI4肝纤维化大鼠对肝组织转化生长因子β1 (transforrning growing factor TGF-β1)、间质胶原蛋白(COL)Ⅰ、Ⅲ表迭的影响。方法以CCl4复合因子刺激8周刺备Wistar大鼠肝纤维化模型后,予丹芍化纤胶囊治疗8周,收集血、尿及肝组织标本, 测定血尿羟脯氨酸(Hyp)、血清谷丙转氨酶(ALT)及透明质酸(HA);观察肝组织纤维化程度、TGF- β1及COL Ⅰ、Ⅲ的表达水平。结果模型组、自然恢复组及治疗组动物肝组织出现不同程度肝纤维化病变,TGF-β1和COL Ⅰ、Ⅲ表达水平升高,其中模型组各指标升高最明显。丹芍化纤胶囊治疗组各项指标虽高于正常对照组,但与模型组比较有明显改善。结论丹芍化纤胶囊可能通过降低肝组织TGF-β1的表达水平,减少细胞外基质中COLⅠ、Ⅲ的沉积,同时通过促进其降解而起到治疗肝纤维化的作用。 Objective To investigate the effects of the traditional Chinese medicine that Dan-Shao-Hua-Xian(DSHX) capsule on the expression of TGF-β1(transforming growing factor β1) and the expression of collagen Ⅰ 、Ⅲ in rat hepatic fibrosis. Methods Male Wistar rats were induced by subcutaneous injection CCl4, drinking alcohol, giving diet of high-fat and hypoprotein for producting hepatic fibrotic models for 8 weeks. Then experimental rats were treated by DSHX for 8 weeks. At the end of the experiments, the samples of blood, urine and liver were collected to examine the urinal hydroxyproline(Hyp).serumal alanine aminotransferase(ALT), serurnal hyaluronic acid(HA) and the degree of hepatic fibrosis. The expression of TGF-β1 and COL Ⅰ .Ⅲ were evaluated by immunohistochemical techniques. Results Compared to the normal control group, the stage of hepatic fibrosis.levels of set- tam HA、ALT and the urinary excretion of Hyp were increased significantly in CCl4-induced hepatic fi- brosis group. The expression of TGF-β1 and COLⅠ、Ⅲ also increased in CCl4-induced hepatic fibrosis group 、non-DSHX-treated group and treated group. Although every item of DSHX treated group were increased than the normal control, it was significantly decreased when compared to CCl4-induced hepatic fibrosis group. Conclusion The DSHX capsule could inhibit the expressions of TGF-β1 and COL Ⅰ、Ⅲ in the liver tissues of CCl4-induced hepatic fibrosis rats, which might be related with its anti-hepatic fibrosis activity.
