摘要
目的:探讨黄芪对肾间质纤维化改善的作用机制。方法:体外培养人肾间质成纤维细胞(KFB),传至3代后分为3组:对照组(常规培养),血管紧张素Ⅱ(AngⅡ)培养组(在培养液中分别加入10-8mol/L、10-7mol/L和10-6mol/L的AngⅡ),黄芪注射液干预组(分别于培养液中加入10-6mol/L的AngⅡ和10mg/L、20mg/L和40mg/L黄芪注射液)。作用48h后,各组用MTT比色法测定细胞增殖情况,用ELISA法测定细胞培养上清液中TGF-β1的水平。结果:AngⅡ可促进KFB增殖,刺激TGF-β1的分泌,且呈剂量依赖性(r分别为0.612和0.723,P<0.05);黄芪注射液可抑制AngⅡ的上述作用,且呈剂量依赖性(r分别为-0.561和-0.689,P<0.05)。结论:黄芪可以通过抑制成纤维细胞增殖和TGF-β1的分泌,延缓肾间质纤维化进展。
Aim: To explore the protective effects of astragalus membranaous on human renal interstitial fibroblasts. Methods: The human renal interstitial fibroblasts were cultured. The third generation cells were allocated into three groups: control group( cells were cultured under normal conditions) , Ang Ⅱ groups ( cells were cultured with 10^-8 mol/L, 10^-7 mol/L,or 10^-6 mol/L Ang Ⅱ ) , Astragalus membranaous groups( cells were cuhared with 10^-6 mol/L AngⅡ and 10 mg/L,20 mg/L, or 40 mg/L astragalus membranaous) , respectirely. After 48 h, the cellular proliferation was determined using MTT colorimetry, the levels of TGF-β1 in the supernatants were measured using ELISA. Results: AngⅡ induced the proliferation and the secretion of TGF-β1 in fibroblasts in dose dependent way, while astragalus membranaous reduced the effects of Ang Ⅱ. Conclusion: By inhibiting the proliferation and the secretion of TGF-β1 in fibroblasts, astragalus membranaous can delay the progression of renal interstitial fibrosis.
