摘要
用含4096点的cDNA芯片研究单纯低剪应力(4.20dync/cm2,2h)作用下或生理浓度17β雌二醇孵育48h后再经相同条件的低剪应力作用后内皮细胞基因的差异表达。结果发现:与未行剪应力处理的对照组相比,单纯低剪应力组共有108条基因(53条基因表达增加,55条基因表达下降)出现差异表达,约占芯片上基因总数的2.6%。其中抑癌基因、与脂质代谢的有关基因明显上调,而与应激反应、细胞的代谢的有关基因的表达则明显降低。先用雌激素孵育再行低剪应力处理的内皮细胞与未经任何处理的对照组比较,共有384条基因(226条基因表达增加,158条基因表达下降)出现差异表达,约占芯片基因总数的9.4%。在表达上调的基因中其转录产物涉及PGI2合成、单核巨噬细胞的吞噬作用等;在表达下调的基因中其转录产物涉及纤维蛋白原的降解、原癌基因产物的生成等。
cDNA microarray technology is a powerful and efficient tool to rapidly compare the differential expression of a large number of genes. We employed this approach in this article to analyze gene expression profiles of cultured human umbilical vein endothelial cells (HUVECs) exposed to low fluid shear stress (4.20dyne/cm^2 ) for 2h or incubated with 17β-estradiol for 48h followed by being exposed to low fluid shear stress. Normal static cultured HUVECs were selected as a control. Total RNA from normal static cultured HUVECs was labeled by Cy3-dCTP and total RNA of HUVECs exposed to low shear stress or incubated with 17β-estradiol( 10^-7 mol/L) followed by being exposed on low shear stress was labeled by Cy5-dCTP. After bioinformatics analysis, among the 4096 target gene, a total of 108 genes (approximately 2.6% ) revealed differential expression in HUVECs exposed to low shear stress,53 genes were up-regulated and 55 were down-regulated. In HUVECs incubated with 17β-estradiol followed by being exposed to low shear stress, 384 genes (approximately 9.4% ) revealed differential expression, 226 genes were upregulated and 158 were down-regulated.
