人脐血单个核细胞透过血脑屏障的可能性 被引量:4

摘要 目的:探讨血脑屏障对脐血单个核细胞的选择性通透性。资料来源:应用计算机检索Medline和Elsevier全文数据库1996/2004与脐血单个核细胞及血脑屏障相关文章,检索词“umbilicalcordbloodcells,mononuclearcells,mesenchymalstemcells,Intravenous,Intraarterial,intracarotid,bloodbrainbarrier”,并限定文章语言种类为English。同时计算机检索中国期刊网专题全文数据库2002/2004期间的文章,检索词“脐血细胞、单个核细胞、间充质干细胞、血脑屏障”,限定文章语言种类为中文。资料选择:对资料进行初审。纳入标准:有关血脑屏障和脐血单个核细胞或间充质干细胞诱导分化、向脑实质定向迁移的文献。排除标准:文献中重复研究、综述类文章。资料提炼:共收集到38篇相关文献,其中10篇因系重复研究而被筛除。选取的28篇分别选自19种不同的杂志。资料综合:28篇文献对脐血单个核细胞如何透过血脑屏障进入脑实质进行综合分析。研究表明,移植脐血单个核细胞能够透过血脑屏障在宿主中枢神经系统内存活、分化为神经细胞,并迁移到病变部位与宿主神经细胞在形态结构和功能上形成整合而改善感觉、运动和认知等神经功能的缺损。不同的病理生理情况下血脑屏障对其有选择性通透作用,脐血单个核细胞进人脑组织的主要机制为黏附分子介导的特异性转移、血脑屏障固有簿弱环节、血脑屏障通透性的改变及病变组织的趋化作用等。结论:已证实脐血单个核细胞经外周血管输入可透过血脑屏障,并改善神经功能的缺损,这为移植干细胞治疗中枢神经系统疾病提供了理论基础。 OBJECTIVE: To review the permeability of mononuclear cells derived from human umbilical cord blood cells passing blood brain barrier. DATA SOURCE: By using computer, the related articles were searched for from MEDLINE database and the Elsevier full text database between 1996 and 2004 with key words of “umbilical cord blood cells, mononuclear cells, mesenchymal stem cells, intravenous, intraarterial, intracarotid, blood brain barrier”, and the language was limited to English. Meanwhile, the Chinese articles were searched for from the Chinese journals full text database between 2002 and 2004 with the key words of “umbilical cord blood cells, mononuclear cells, mesenchymal stem cells, blood brain barrier”, and the language was limited to Chinese. STUDY SELECTION: The articles were browsed firstly. Inclusion criteria were for these articles about blood brain barrier and differentiation and migration into brain of mononuclear cells or mesenchymal stem cells derived from human umbilical cord blood cells. Exclusion criteria were for those about repetitive studies and reviews. DATA EXTRACTION: Totally 38 related literatures were collected, of which 10 articles were, excluded for the repeating research. The rest 28 articles were selected from 19 journals. DATA SYNTHESIS: How mononuclear cells derived from human umbilical cord blood cells to pass blood brain barrier and migrate into brain was summarized and analyzed in 28 articles. It suggested that the transplanted mononuclear cells of umbilical cord could survive in host central nervous system through blood brain barrier, differentiate into nerve cells, and migrate to the site of lesion. They were integrated with host nerve cell on formation and function to improve defect of neural function, including sensation, motor and cognition. Various permeabilities were selected at different pathophysiological conditions. The main mechanism of mononuclear cells of umhilieal cord blood going into human cerebral tissue was mediated specific transfer of adherent molecules, inherent weak link of blood brain barrier, changes of permeability of blood brain barrier and changes of ehemotaxis of lesion tissue, CONCLUSION: It is proved that mononuclear cells derived from human umbilical cord blood cells can pass blood brain barrier and improve defect of neural function, which provides a themry for the treatment of central nervous system disease with transplantation of stem cell.
