摘要 目的探讨亚砷酸对人肝癌细胞BEL-7402凋亡及ERK-1蛋白表达的影响.方法通过体外细胞培养,用流式细胞术观察BEL-7402细胞周期及凋亡率变化;HE染色法、荧光显微镜观察细胞的凋亡形态变化;并用免疫组化法检测细胞ERK-1蛋白的表达.结果亚砷酸(1.0~8.0 μmol/L)能诱导BEL-7402细胞凋亡并阻滞细胞周期于S、G2/M期,呈剂量依赖性;亚砷酸(8.0 μmol/L)作用BEL-7402细胞48 h后, HE染色、荧光显微镜观察可见BEL-7402细胞呈现明显的凋亡形态改变;免疫组化法检测发现8.0 μmol/L的亚砷酸作用48 h 后BEL-7402细胞的ERK-1蛋白表达明显减弱. 结论亚砷酸体外有诱导人肝癌细胞凋亡及降低ERK-1蛋白表达的作用. Objective To investigate the effects of arsenious acid on the apoptosis and the expression of ERK-1 protein in HCC cell line BEL-7402. Methods The cells were cultured in vitro, and the cell cycle and apoptosis rate were detected by flow cytometry, The morphologic changes of cell apoptosis were observed by HE staining and fluorescnce microscope,and the expression of ERK-1 protein was determined by immunohistochemical method. Results Arsenious acid in concentrations from 1.0 μmol/L to 8.0 μmol/L could induce the cell apoptosis and arrest the cells at S,G2/M phase in a dose dependent manner. After treatment with 8.0 μmol/L arsenious acid for 48 h, BEL-7402 cells showed evident morphologic .changes of apoptosis; and in cells treated by arsenious acid, ERK-1 expression was significantly down-regulated at protein level. Conclusion In vitro, arsenious acid can induce the apoptosis and lower expression of ERK-1 protein of human hepatocarcinoma cells.
