黄芪对急性弓形虫感染小鼠的血清IFN-γ及IL-18的影响 被引量:4

摘要 目的研究黄芪对RH株弓形虫100个速殖子感染的小鼠血清IFN-γ及IL-18水平的动态影响,以探讨黄芪对急性弓形虫感染小鼠保护作用的机制。方法建立102速殖子感染小鼠模型,设黄芪治疗组和对照组。两组分别定期处死小鼠取血清。用ELISA测定感染后2、4、6、8d血清IFN-γ水平及感染后4、6、8d的血清IL-18水平。结果感染后黄芪组IFN-γ水平持续上升,4d和8d分别为21.0pg/ml和15.36pg/ml,显著高于对照组(5.62pg/ml和9.73pg/ml)(P<0.05)。感染后黄芪组血清IL-18水平持续上升,8d为594.75pg/ml,显著低于对照组(709.00pg/ml)(P<0.05)。结论黄芪可能通过双向调节宿主的非特异免疫应答对急性弓形虫感染小鼠产生保护作用。 Objective To evaluate the dynamic impact of Astragalus membranaceus on the levels of IFN-γand IL-18 in sera from mice infected with 100 tachyzoites of virulent RH strain of Toxoplasma gondii, and to study the mechanism of protective effect of Astragalus membranaceus on the mice with acute toxoplasmosis. Methods Mice infected with 102 tachyzoites were administered orally with Astraglus membranaceus and the control group was given water simultaneously. Both the groups of mice were killed every other day to collect sera. The serum levels of IFN-γ on the 2,4,6 and 8 dpi(day post infection) and IL-18 on the 4,6 and 8 dpi were assayed by ELISA. Results Before 6 dpi the serum levels of IFN-~ in both groups kept elevating with the level of 21.0 pg/ml and 15.36 pg/ml at 4 dpi and 8 dpi in Astraglus group,which were significantly higher than those in control group (5.62 pg/ml and 19.73 pg/ml) (P 〈0. 05). The serum levels of IL-18 in both groups kept elevating with the level of 594.75 pg/m at 4 dpi in Astraglus group, which was significantly lower than that of control group (709.00 pg/ml ) ( P 〈 0.05 ). Conclusion The curves of serum levels of IFN-γ/and IL-18 in Astraglus group change in a more relax way than those in control group, which indicates the protection of Astraglus against Toxoplasmosis is through its deliberate regulation of immune responses of hosts.
