大鼠单侧隐睾对侧睾丸生精细胞凋亡与T-AOC、MDA和Bcl-2基因表达的关系 被引量:3

摘要 目的探讨大鼠单侧隐睾对侧睾丸生精细胞凋亡发生机制。方法将SD雄性大鼠20只随机分为实验组(10只)和对照组(10只),建立单侧隐睾大鼠模型。术后90d取手术对侧睾丸,采用TUNEL法检测生精细胞凋亡,免疫组化SP法检测Bcl-2基因表达,化学比色法测定睾丸组织中总抗氧化能力(T-AOC)和丙二醛(MDA)的含量。结果实验组手术对侧睾丸重量减轻,生精细胞凋亡指数(AI)增高,Bcl-2基因表达降低,总抗氧化能力下降,丙二醛含量增高,与对照组相比,两组差别具有显著性(P<0.01)。结论大鼠单侧隐睾可导致对侧睾丸生精细胞过度凋亡,生精功能严重受损,致使生育能力下降。 Objective To investigate germ cell apoptosis and apoptotic mechanisms in the contralateral testes of unilateral experimental cryptorchid rats. Methods Immature rats(22 day-old Sprague-Dawley) were subjected to unilateral cryptorchidism.Germ cell apoptosis was detected by TdT-mediated dUTP-x nick end labeling (TUNEL). The expression of Bcl-2 were studyed by immunohistochemical method. Biochemical parameters (T-AOC,MDA) were evaluated by specreophotometric method. Results Compared with control group, the number of apoptotic germ cells in the cryptorchid group was increased significantly, but its testes weight was decreased predom icantly.The contents of MDA was significantly higher than in the control, the level of T-AOC and Bcl-2 was predominantly lower. Conclusion The levels of T-AOC,MDA and Bcl-2 might be involved in the germ cell apoptosis in the contralateral testes of experimental unilateral cryptorchid rats
