摘要
从养殖场发病家禽中分离到多株禽源性大肠杆菌(E scherich ia coli),用其中Y ZG 040515、NTLFC 040402、CHZ031016菌株提取细菌高致病性染色体DNA,根据耶尔森菌高致病性毒力岛(HP I)irp 2基因设计引物,用PCR扩增出278 bp基因片段。将PCR产物克隆到pGEM-T-easy中,经E coRⅠ酶切鉴定为阳性者进行序列测定。结果表明:该基因片段与G eneB ank中发布的耶尔森菌HP I irp 2基因的同源性高达98%以上,证明禽源性大肠杆菌中具有耶尔森菌毒力岛HP I基因序列,结合临床发病情况和流行病学,提示该毒力岛与日趋严重的家禽大肠杆菌病可能有一定的相关性。
Avian Escherichia coli strains were isolated from chicken with colibacillosis. The chromosome DNAs of E. coli were extracted. A cDNA fragment was amplified by PCR with primers designed according to the sequence of the Yersinia high-pathogenicity island (HPI) irp2 gene published in Genbank. All of 3 PCR products were cloned into pGEM-T-easy and sequenced. The results showed that the sequences from 3 E. coli strains shared more than 98% homology with the irp2 gene of Yersinia HPI. It indicates that the HPI has existed in avian E. coli strains and it may be related to the pathogenicity of poultry E. coli in poultry.
