摘要
目的:建立用离子抑制-反相高效液相色谱快速测定池杉、落羽杉根系中 L-苹果酸、莽草酸含量。方法:采用 Agilent1100 DAD 检测器,Hypersll ODS2 5 μm(4.6mm×150mm)柱,HClO_4(pH=2.5)溶液为流动相,柱温30℃时于214 nm 波长处以外标法测定。结果:L-苹果酸、莽草酸分别在7.16×10^(-6)mol·L^(-1)2.24×10^(-2)mol·L^(-1)和1.27×10^(-7)mol·L^(-1)~2.06×10^(-4)mol·L^(-1)范围内,浓度与峰面积有良好的线性关系(r=1.0000和r=0.9999)。检测限分别为2.42×10^(-8)mol·L^(-1)和1.16×10^(-9)mol·L^(-1)(S/N=3)。结论:该法简便、快速、有效、重现性好,首次用于实际池杉、落羽杉根系中 L-苹果酸、莽草酸的测定,取得了很好的结果。
Objective :A ion - suppression reversed - phase HPLC method was established for the determination of rudimental L - malic acid and Shikimic acid in Taxodium ascendems and T axodium distichum. Methods:They can be separated on Agilent 1100 DAD and Hypersil ODS25μm(4.6mm×150mm)column by using perchloric acid ( pH = 2.25 ) as mobile phase with UV detector at a wavelength of 214 nm. Results: The standard curve of L - malic acid and shikimic acid was linear in the concentration range 7.16 ×10^-6mol· L^-1 -2.24 ×10^-2mol· L^-1 and 1.27 ×10^-7mol· L^-1 -2.06 ×10^-4mol· L^-1 and the correlation coefficient was 1. 0000 and 0. 9999. The detection limit was 2.42 ×10^-Smol· L^-1 and 1.16 ×10^- 9mol· L^-1 (S/N =3). Conclusions:The method has been successfully applied to the determination of trace rudimental L - malic acid and Shikimic acid in the root of Taxodium ascendens and Taxodium distichum,which was proved to be accurate, simple effective and feasible
