摘要 目的探讨Survivin基因在结直肠癌中的表达及其意义。方法应用免疫组织化学(SP)法和TdT介导的dUTP缺口末端标记技术原位观察48例结直肠癌患者的癌组织、癌旁组织和正常组织中Survivin蛋白的表达和细胞凋亡。结果癌组织Survivin的阳性表达相对含量(PU)值(38.76±5.14)明显高于癌旁(25.17±7.26)和正常组织(0.57±0.03),差异有统计学意义(t=2.54,P<0.05),正常大肠黏膜中极少见Survivin蛋白表达;癌旁和癌组织凋亡指数(AI)分别为(7.51±2.63)%和(4.65±1.76)%,两者差异有统计学意义(t=2.18,P<0.05);Survivin蛋白的阳性表达与患者的性别、年龄、肿瘤大小无相关性(P>0.05),与癌组织的分化程度、淋巴结转移和Dukes分期相关,低分化、有淋巴结转移和DukesC/D期的Survivin蛋白表达显著高于相关组别(P<0.05)。结论Survivin在结直肠癌组织中表达上调,Survivin表达可抑制结直肠癌的细胞凋亡,这可能是结直肠癌发生、发展的有关因素;Survivin的表达与结直肠癌的恶性生物学行为有关。 Objective To investigate the role of Survivin expression in the pathogenesis of colorectal carcinoma. Methods Immunohistochemistry S-P method and terminal deoxynudeotidyl transferase-mediated dUTP nick end labeling (TUNEL) were used to detect the expression of Survivin and apoptotic cell in situ in colorectal cancerous tissues, paracancerous tissues and normal tissues from 48 cases .Results The Survivin positive unit (PU) was higher in cancerous tissues (38.76 ± 5.14) than in paracancerous (25.17 ± 7.26) or normal tissues (0.57 ± 0.03) ( t = 2.54, P 〈 0.05). The apoptosis index (AI) of paracancerous tissues was (7.51 ± 2.63)%, higher than cancerous tissues (4.65 ± 1.76)% (t = 2.18, P 〈 0.05). The higher expression of Survivin was related to pathological grade, lymph node and Dukes stage. Conclusion Survivin expression may play an important role in carcinogenesis of colorectal carcinoma and be associated with reduced AI.
