三氧化二砷对人脑胶质瘤生长的抑制作用 被引量:9

Inhibitory effect of arsenic trioxide on the growth of human TJ905 glioblastoma cells

摘要 目的观察三氧化二砷(As2O3)对人脑胶质瘤细胞增殖抑制作用,探讨以As2O3治疗胶质瘤的可行性。方法将不同浓度As2O3(浓度分别为:0、0.5、1、2、3和4μmol/L)分别加入体外培养的TJ905人多形性胶质母细胞瘤,通过噻唑蓝(MTT)比色法检测体外细胞增殖率、PCNA染色检测细胞增殖活性、TUNEL法检测原位细胞凋亡和Western印迹检测bc12蛋白表达水平。结果与对照组相比,治疗组TJ905细胞增殖明显下降,且各组差异均有统计学意义(P<0.01),TUNEL法表明细胞凋亡指数上升(P<0.01);Western印迹显示凋亡相关基因bcl2蛋白表达水平呈下降趋势。结论As2O3具有抑制人脑恶性胶质瘤细胞增殖的作用,可能成为临床治疗胶质瘤的候选化疗药物之一。 Objective To explore the inhibitory effect of arsenic trioxide (As203) on the growth of human TJ905 gliohlastoma cells (TJ905 cells) and the feasibility of using As203 treating gliomas. Methods TJ905 cells were treated by different doses of As203(0, 0,5, 1, 2, 3 and 4μmol/L). MTT assay and staining for PCNA were used for cell proliferation, and TUNEL method for determination of cell apoptosis respectively. Western blot was applied to detect the bcb2 expression. Results All the treated cells showed decreased proliferation in vitro as detected by MTT method and staining for PCNA, The proliferation rate between treated group and control group was significantly different (P 〈 0.01). In situ labeling apoptotic DNA fragment of the treated cells demonstrated that the cell apoptosis was significantly increased following treatment with As203 (P 〈 0.01), Western blot revealed the expression of hcl-2 prorein was decreased. Conclusion As203 can inhibit the proliferation of human TJ905 cells, suggesting that As203 may be a good candidate for chemotherapy of human gliomas,
