摘要
目的观察甲基泼尼松龙对周围神经损伤修复后的形态学变化。方法采用45只成年SD雄性大鼠,取双侧股外侧切口,钳夹双侧坐骨神经造成神经损伤,即刻于神经损伤局部肌肉间隙内给药,根据药物不同分为5组。大剂量甲基泼尼松龙组:每侧剂量为30mg;小剂量甲基泼尼松龙组:每侧15mg;地塞米松组:每侧2mg;对照组:用等量生理盐水;及正常组。术后4周取损伤神经远端神经,作成横切片后锇酸染色,计数轴索总数目及单位视野再生髓鞘数目,并行统计学分析。结果腓总神经轴索总数目:大剂量甲基泼尼松龙组及地塞米松组优于对照组(P<0.05);小剂量甲基泼尼松龙组明显优于对照组(P<0.01),与正常组相比差异无统计学意义(P>0.05)。单位视野有髓神经纤维数目:大、小剂量甲基泼尼松龙组均明显优于地塞米松组及对照组(P<0.01),与正常组相比差异无统计学意义(P>0.05)。结论神经损伤后局部应用甲基泼尼松龙可起到促周围神经再生的作用。
Objective To investigate the effect of topically applied methylprodnisolone (MP) on the repair of peripheral nerve injury by histological study. Methods 45 male SD rats were randomly divided into 5 groups. Sciatic nerve injury model was established by clamping the nerve on both sides. Various dosages of MP were applied to the injury sites according to group assignment, namely 30mg MP/side, 15mg MP/side, 2mg dexamethasone/side, and 0.4ml normal saline/side. Rats without sciatic nerve injury served as control. Evaluation was carried out 4 weeks after the surgery. The nerves distal to the injury site were harvested for osmie acid staining. The total axon numbers and axon numbers per area were calculated. Results The total axon numbers of the two MP groups and the dexamethasone group were significantly higher than that of the control group ( P 〈 0.05 ). The average numbers of myelinated nerve fibers per area of the two MP groups were significantly higher than those of the dexamethasone group and the control group ( P 〈 0.01 ). Conclusion Topical application of MP can facilitate the repair of peripheral nerves injury in rats.
