丹参对成骨细胞株MC3T3─E1细胞碱性磷酸酶活性及DNA合成影响的实验研究 被引量:15

摘要 选择克隆化成骨细胞株——MC3T3-E1细胞,采用细胞培养方法,观察细胞内DNA合成与碱性磷酸酶(ALPase)活性,了解丹参对体外培养的成骨细胞生长、分化与代谢的影响。研究结果显示,丹参明显增强MC3T3-E1细胞内ALPase活性,5.0mg/ml浓度时,丹参对MC3T3-E1细胞内碱性磷酸酶活性影响最大,比对照组增强约135%。丹参这种促进作用只限于分化晚期的MC3T3-E1细胞,而对分化早期细胞内碱性磷酸酶活性产生抑制作用。其次,这种作用还与药物作用时间有关。但是,丹参对各生长分化期的MC3T3-E1细胞内DNA合成均无影响。本研究结果表明,丹参有明显增强体外培养分化晚期的MC3T3-E1细胞内碱性磷酸酶活性的作用,这种作用不是通过促进细胞增殖,而是改善细胞功能实现的,并且受丹参药物浓度与作用时间的影响,说明丹参可能在促进正畸牙齿移动中新生牙槽骨形成方面发挥作用。 The authors previously found that SMB could increase the rate of remodel of periodontium and accelerate the orthodontic tooth movement in rabbits. To understand the role of SMB on the metabolism of cells in remodeling of peridontium, authors studied the effects of SMB on DNA synthesis and alkaline phosphatase (ALPase) activity of the cloned osteoblast like cells MC 3T3 E1. The cells were cultured in a MEM with 0.3% of fetal bovine serum and treated with SMB at the concentration of 0.1 mg/ml ̄10.0 mg/ml. There was no difference in DNA synthesis between the groups in different dosage of SMB and the group of control. On the other hand, SMB increased ALPase activity in a dose dependent fashion in multilayer stage of cells, and up to maximum at the level of 5.0 mg/ml, in which concentration ALPase activity was about 135% greater than that of control. However, ALPase activity was inhibited in monolayer stage of cells by the addition of SMB. These results suggest that SMB has the stimulating effect on the phenotype of osteoblast like cells in vitro, and may play an important role on accelerating the remodel of bone in vivo as well.
