摘要
新生猪胰腺经胶原酶消化,分离纯化得纯净胰岛。用BaCl_2交联一步法微囊技术包裹该胰岛,所得微囊直径500~700μm,每个微囊含1~3个胰岛。移植治疗Ⅰ型糖尿病5例,每例移植胰岛数为13±2万,术后不使用免疫抑制剂。受体血糖移植前为18.2±3.74mmol/L,用药后降至8.14±2.06mmol/L,血清C肽由113.5±58pmol/L升至247±83pmol/L。外源胰岛素用量显著减少,从移植前的64±15.48U/d减至27.2±9.44U/d。提示采用微囊化新生猪胰岛异种移植,能有效地纠正患者高血糖,显著减少胰岛素用量。证明海藻酸钠微囊具有良好的免疫隔离作用和生物膜性能,可有效地避免免疫排斥反应。
The pancreata of new born pigs were digested by collagenase and then isolated to collect pure islets,these islets were prepared to microcapsules by Bariumcross linked one-step method with alginate. The diameter of microcapsules is 500-700μm, and every capsule cantains 1-3 pancreatic islets. The capsules were transplanted to five IDDM patients without immunodepressive. After transplantation fasting blood sugar of the patients decreased from 18.2± 3.74mmol/L to 8.14 ±2.06mmol/L. The dosage of injection insulin reduced from 64±15- 48U/d to 27.2 ± 9.44U/d, serum C peptide increased from 113.5± 58pmol/L to 247±83pmol/L. The curative effect was maintained for more than a hundred days.The results reveals that the graft of microencapsulated pancreatic islets effectively correct the blood sugar of IDDM. It also shows that themicroencapsulation had the immunoisolation and biomembrane function, it can protect the host from antigraft rejection.
