用精原干细胞移植技术制作转基因动物的研究前景 被引量:5

摘要 在生物科学、生物医学、畜牧业科学和生物反应器的研究中,转基因动物是非常有用的工具.然而,最常用的制作转基因动物的方法,如原核显微注射和核移植效率都非常低.在最近10年中,研究人员利用精原干细胞移植技术发明了一系列新方法.其中的一些方法一旦与遗传修饰方法相结合,就会形成一套制备转基因动物的新方法.本文综述了与转基因动物制作相关的精原干细胞移植研究成果,如精原干细胞移植技术、精原干细胞的冷冻保存方法、精原干细胞移植受体的准备、精原干细胞的长期增殖和传代培养、精原干细胞的遗传修饰及外源基因的遗传特性等,描述了用精原干细胞移植技术制作转基因动物的一整套方法.根据所用供体和受体的不同,将这种新的转基因动物制作方法分为两类:同种移植法和同体移植法.这一研究领域的进展虽然迅速,但是每一种方法仍然有潜在的困难有待于克服.本文对这些新方法的优点和目前存在的问题进行了探讨.
