摘要 目的:观察成纤维细胞生长因子对软骨细胞增殖和细胞外基质合成的影响。方法:实验于2003-03/2004-12在第四军医大学口腔颌面外科实验室和南方医科大学南方医院组织工程中心完成。酶消化法从兔耳获取软骨细胞与12g/L藻酸钠混合,细胞浓度1010L-1,经注射器滴入125mmol/L的氯化钙溶液,形成软骨细胞/藻酸钙凝胶微球,用含体积分数为0.1胎牛血清的DMEM培养液体外培养,培养液中分别加入0,10,50和100μg/L浓度的碱性成纤维细胞生长因子,14d终止培养,进行生物化学分析。观察各组DNA总量、糖胺多糖总量、单位DNA的糖胺多糖含量。结果:①DNA总量:0,10,50和100μg/L成纤维细胞生长因子浓度组分别为(3.44±0.23),(4.70±0.22),(6.60±0.35)和(6.82±0.33)μg。②糖胺多糖总量:0,10,50和100μg/L成纤维细胞生长因子浓度组分别为(15.35±0.80),(20.63±0.72),(29.72±2.08)和(31.11±2.39)μg。③单位DNA的糖胺多糖含量:0,10,50和100μg/L成纤维细胞生长因子浓度组分别为(4.47±0.09),(4.39±0.13),(4.50±0.13),(4.55±0.18)g/g。结论:成纤维细胞生长因子明显促进了软骨细胞的增殖,糖胺多糖合成总量也得到明显提高,但单位DNA的糖胺多糖合成量无明显变化,表明成纤维细胞生长因子影响软骨细胞的生物学特性。 AIM: To observe the effects of fibroblast growth factor on the cartilage cell proliferation and synthetic extracellular matrix. METHODS: The experiment was conducted at the Laboratory of Oral and Maxillofacial Surgery, Fourth Military Medical University and Center of Tissue Engineering, Southern Hospital, Southern Medical University between March 2003 and December 2004, The chondrocytes were isolated from rabbit auricular cartilage by enzyme digestion and mixed with 12 g/L alginate, with the cell concentration of 10^10 L^-1, and the 125 mmol/L CaCl2 solution was dropped by injection syringe to prepare the alginate beads of chondrocytes and cultured with DMEM medium of foetus ox serum with the volume fraction of 0,1 in vitro added with 0, 10, 50 and 100 μg/L basic fibroblast growth factor (bFGF) for 14 days to performed the biochemical analysis. The total amount of DNA and glycosaminoglycan and the content of glycosaminoglycan of unit DNA were observed in every group. RESULTS: ① The total amount of DNA:It was(3.44±0.23), (4.70±0.22), (6.60±0.35) and (6.82±0.33)μg, respectively in the 0,10,50 and 100μg/L fibroblast growth factor concentration group. ② Total amount of glycosaminoglyean: It was (15.35±0.80), (20.63±0.72), (29.72±2.08) and (31.11±2.39) μg, respectively in the 0,10,50 and 100 μg/L fibroblast growth factor concentration group. ③ Content of glyeosaminoglyean of unit DNA: It was (4.47±0.09), (4.39±0.13), (4.50±0.13) and (4.55±0.18) g/g, respectively in the 0,10,50 and 100 μg/L fibroblast growth factor concentration group. CONCLUSION: The fibroblast growth factor accelerates significantly the proliferation of ehondrocytes, and the total content of synthetic glyeosaminoglyean also enhances obviously, but the synthetic amount of glyeosaminoglyean of unit DNA has insignificant changes. It is indicated that fibroblast growth factor affects the biological character of the chondrocytes.
