摘要
用尸体头皮经无菌处理后剪成０．３～０．５ｃｍ宽的皮条，再从真皮皮下组织交界处剪开，从皮下组织中轻轻拔出毛囊，培养于加有胰岛素、氢化可的松的１０％新生牛血清ＤＭＥＭ培养基中，结果显示游离毛囊前２天生长最快，前４天平均生长长度为０．１７ｍｍ／ｄ。胰岛素和氢化可的松对维持毛囊的形态有作用。放射自显影显示ＤＮＡ的合成主要是发生在毛母质细胞和部分外根鞘的基底细胞中，表明毛囊的生长延长是由于毛母质细胞分裂、增生的结果，外根鞘细胞能参与组织的修复。培养４天后毛囊出现贴壁，角朊细胞从外根鞘中迁出，一般７～１０天都会贴壁，前４天毛囊的形态没有明显的改变，以后毛根变成棒状，毛乳头紧凑成一团。
Folowingsterilizationhumanscalpswerecutintothinstripsabout0.3～0.5cmwide,thentheskinwasseparatedatthedermo-subcutaneousfatinterface.Theintacthairfolicleswerepuledoutfromthesubcutaneousfatusingwatchmakers'forcepsandmaintainedfree-floatinginDMEMmediumsupplementedwithinsulin,hydrocortisoneand10%new-borncalfserum.Theyshowedasignificantincreaseinlengthover4days(P<0.01),theaveragerateofgrowthwas0.17mm/dandquickestinthefirsttwodays.3H-thymidineautoradiographyshowedthatDNAsynthesismainlyoccurredinthehairmatrixcelsandsomebasalcelsofouterrootsheath,whichindicatedthatthegrowthandprolongationofhairfolicleswereresultedfromthedivisionandproliferationofhairmatrixcels,andsomebasalcelsofouterrootsheathtookpartinthegrowthofouterrootsheath.Af-terfourdaysofculturethehairfoliclesbegantoatachandkeratinocytesmigratedoutfromouterrootsheath.Inthefirstfourdaysthemorphologyofhairfoliclesremainedunchanged,thenthehairshaftbecameclubshaped,andthedermalpapilabecameatightbalofcels.
