摘要
用尸体头皮经无菌处理后剪成0.3~0.5cm宽的皮条,再从真皮皮下组织交界处剪开,从皮下组织中轻轻拔出毛囊,培养于加有胰岛素、氢化可的松的10%新生牛血清DMEM培养基中,结果显示游离毛囊前2天生长最快,前4天平均生长长度为0.17mm/d。胰岛素和氢化可的松对维持毛囊的形态有作用。放射自显影显示DNA的合成主要是发生在毛母质细胞和部分外根鞘的基底细胞中,表明毛囊的生长延长是由于毛母质细胞分裂、增生的结果,外根鞘细胞能参与组织的修复。培养4天后毛囊出现贴壁,角朊细胞从外根鞘中迁出,一般7~10天都会贴壁,前4天毛囊的形态没有明显的改变,以后毛根变成棒状,毛乳头紧凑成一团。
Folowingsterilizationhumanscalpswerecutintothinstripsabout0.3~0.5cmwide,thentheskinwasseparatedatthedermo-subcutaneousfatinterface.Theintacthairfolicleswerepuledoutfromthesubcutaneousfatusingwatchmakers'forcepsandmaintainedfree-floatinginDMEMmediumsupplementedwithinsulin,hydrocortisoneand10%new-borncalfserum.Theyshowedasignificantincreaseinlengthover4days(P<0.01),theaveragerateofgrowthwas0.17mm/dandquickestinthefirsttwodays.3H-thymidineautoradiographyshowedthatDNAsynthesismainlyoccurredinthehairmatrixcelsandsomebasalcelsofouterrootsheath,whichindicatedthatthegrowthandprolongationofhairfolicleswereresultedfromthedivisionandproliferationofhairmatrixcels,andsomebasalcelsofouterrootsheathtookpartinthegrowthofouterrootsheath.Af-terfourdaysofculturethehairfoliclesbegantoatachandkeratinocytesmigratedoutfromouterrootsheath.Inthefirstfourdaysthemorphologyofhairfoliclesremainedunchanged,thenthehairshaftbecameclubshaped,andthedermalpapilabecameatightbalofcels.
