摘要
比较了从土壤中提取DNA的三种方法,结果发现TENS法对不同土壤都有较好的提取效果,提取的DNA片段长度均在23 kbp以上,并且无明显的降解现象,提取效果明显好于SDS高盐提取法和裂解酶法.在DNA纯化过程中,分别比较了试剂盒法、透析袋法和琼脂糖酶法等纯化方法,发现使用琼脂糖酶法纯化粗提的DNA时回收率最高,获得的DNA的质量也很高,纯化DNA可用于酶切等分子生物学操作.
In this paper, three methods of DNA extraction from soil were compared. As a result, the method of TENS is suit for all the soil, the size of DNA fragments is about 23 kbp, and there no degradation. The method of TENS is more efficient than the methods of SDS and lysoenzyme. In purification, DNA was purified with the purification kits, dialysis bag and agarase digest, respectively. Agarase digest is the most efficient of all, which obtains high quality DNA fragments. The soil DNA was then digested by of Sau3AI, and also got the satisfied result.
