摘要
目的观察不同砷化物对原代培养星型胶质细胞的毒性作用。方法细胞活力检测与形态学观察。结果暴露40、60、80、100μmol/L3价和200、300、400、500μmol/L5价无机砷的星型胶质细胞的AlamarBlue还原力与对照组比较差异有显著意义,且呈剂量鄄效应关系。体外24h暴露iAsⅤ和iAsⅢ对原代培养星形胶质细胞的半数细胞活力抑制浓度分别为260.3μmol/L和50.8μmol/L。同时,暴露最低25μmol/L3价和100μmol/L5价无机砷的星型胶质细胞的细胞形态有明显的改变。另一方面,暴露最高1000μmol/L5价甲基砷和二甲基砷的星型胶质细胞的AlamarBlue还原力及细胞形态与对照组比较未见明显差异。结论接触最少25μmol/L的3价无机砷或100μmol/L的5价无机砷对星型胶质细胞可产生明显的毒性损伤作用。3价无机砷的毒性明显大于5价无机砷。在微摩尔浓度范围内的5价甲基砷和二甲基砷对原代培养的星型胶质细胞无明显的直接毒性作用。
Objective To explore whether the toxic effects on the primary cultured astroglia would be induced by different arsenicals. Methods AlamarBlue assay and morphological observation were used to evaluate the toxicity of different arsenicals on astroglia. Results The reduced rates of alamarBlue, which parallel with the viability of cultured cells, in cell cultures treated with 200, 300, 400, 500μmol/L of iAsv or 40, 60, 80, 100μmol/L of iAs^Ⅲ were significantly decreased as compared with the control cell cultures. The 50% effective dose values of iAs^Ⅲ and iAs v to primary cultured astrocytes after 24 h exposure were 260.3 μ mol/L and 50.8μmol/L, respectively. The cytotoxic effects were observed in the cell cultures exposed to 25, 50, 100μmol/L of iAs^Ⅲ and 100, 250, 500μmol/L of iAsv. However, no significant differences were shown between the cell cultures treated with pentavalent methylated arsenicals and control cultures in reduced rates of alamarBlue and morphological observation at the highest levels of exposure. Conclusion Dose-dependent toxic effects in primary cultured astroglia could be induced by exposed to iAsi or iAsv within the concentrations of 25~100μmol/L or 100~500 μmol/L, respectively. Based on the results of this study, the toxic effects induced by iAs^Ⅲ are stronger than those induced by iAsv. On the other hand, no obvious toxic effects on the primary cultured astroglia could be induced by pentavalent methylated arsenicals at micromolar exposed levels.
