摘要
在胶原的羟脯氨酸测定中,传统的酸法水解是在110℃下使用6 mol/L的HCl消解24 h,其操作步骤繁琐且所花时间较长。本文的目的在于通过对酸水解条件的优化,简化羟脯氨酸测定的操作步骤,缩短酸水解的时间。HCl浓度(HC),水解时间(HT)和反应温度(RT)对羟脯氨酸的测定具有显著的影响.酸法水解测定羟脯氨酸的优化条件为:HC=12 N,HT=6 h,RT=110℃。使用优化后的条件测定羟脯氨酸缩短了测定时间,简化了操作,而且灵敏度和重复性较好,可用于快速测定胶原中羟脯氨酸的含量。
In the measurement of the hydroxyproline in the collagen, the traditional acid hydrolysis method is that under 110℃ the animal organism is hydrolyzed with 6 mol/L HCl, for 24 h. The operating steps are very trivial and the hydrolysis time is very long. The purpose of this paper is to simplify the operating steps and shorten the acid hydrolysis time in the measurement of the hydroxyproline by optimization of the acid hydrolysis condition. The HCl concentration ( HC ), hydrolysis time ( HT ) and reaction time ( RT ) have most important affecting for the measurement of the hydroxyproline. The optimum condition of the acid hydrolysis in the measurement of hydroxyproline is: HC =12 mol/L, HT =6 h, RT =110℃. By using the optimum condition in the measurement of the hydroxyproline, the hydrolysis time is shortened and the operation is simplified, and the sensitivity and reproduction are high. This method can be applied to measure the hydroxyproline in the collagen rapidly.
