酚醛树脂作载体固定化葡萄糖淀粉酶的研究

摘要 以酚醛树脂作载体,研究了糖化型淀粉酶固定化条件及固定化糖化酶的性质.结果表明,在 pH=3,T=35℃,t=4h 条件下所得固定化酶活力最高,与游离酶相比,酶活力回收可达82%.糖化酶经固定后催化反应最适 pH=4.2,最适温度为48℃,K_m=28.6mg/mL,装柱连续水解淀粉240h 酶活力保留87%. The phenolic resin was used as carrier to immobilize the glucoamylase.The factors which effect the process of immobilizing were studied using orthogonal test.The best condi- tion of immobilizing is:pH=3,T=35℃,t=4 h and 82% activity of glucoamylase was con- served.The optimum conditions of immobilized glucoamylase action on the substance (starch)are:pH=4.2,T=48℃.K_■=28.6 mg/mL.Continous hydrolysis amylum 240 h.so 87% activity of glucoamylase was conserved.The result shows the immobilized glucoamylase stability on time is high.
