摘要
目的探讨单侧隐睾鼠对侧睾丸生殖细胞和Sertoli细胞变化和还原型谷胱甘肽(GSH)对对侧睾丸的保护作用。方法30只SD雄性大鼠分为对照组(A组)、隐睾组(B组)、隐睾加GSH组(C组),每组各10只。采用化学比色法测定对侧睾丸GSH、丙二醛(MDA)含量;生物素-dUTP/酶标亲和素法检测睾丸生殖细胞的凋亡;透射电镜观察Sertoli细胞的超微结构。结果术后2周,与A组比较,B组对侧睾丸GSH明显降低,MDA和细胞凋亡明显增加(P<0.01),Ser-toli细胞线粒体和滑面内质网扩张。C组这种变化则明显减轻(P<0.01),与A组比较差异无统计学意义(P>0.05)。结论外源性GSH对单侧隐睾鼠对侧睾丸生殖细胞和Sertoli细胞有保护作用。
Objective To investigate the changes of germ and Sertoli cells and the protective effect of glutathione (GSH) in the contralateral testes of unilateral Cryptorchid rats. Methods Thirty male SD rats were divided into 3 groups:control group (A group), cryptorchid group (B group) and cryptorchid + GSH group ( n = 10 in each group). The concentration of GSH and malonic diethyl aldehyde (MDA) in testicular tissues was evaluated by using spectrophotometric method. Germ cell apoptosis was detected by terminal - deoxynudeotidyl transferase mediated d - UTD nick end labeling (TUNEL) method. The ultra- structure of Sertoli cells was observed under transmission electron microscopy. Resuits At 14th day after the operation, as compared with A group, the concentration of GSH in contralateral testes in B group was decreased significantly ( P 〈 0.01 ), as well as the level of MDA and the number of apoptotic germ cell were increased significantly ( P 〈 0.01), and the expansion of mitochondria and smooth endoplasmic reticulum was found in Sertoli cells. But as compared with B group, the above changes were lessened in C group. Conclusion The administration of GSH can protect the germ and Sertoli ceils in the contralateral testes of the rat unilateral cryptorchid.
