摘要
目的观察转化生长因子β1(TGFβ1)及其信号转导蛋白Smad23在去卵巢大鼠骨组织中的表达及意义。方法手术切除大鼠双侧卵巢。术后13周,取离体胫骨测量组织学参数髓腔面积百分率(%Ma.Ar)、骨小梁厚度(Tb.Th),用逆转录多聚酶链反应(RTPCR)检测大鼠骨组织TGFβ1mRNA的表达,用免疫组化法检测Smad23蛋白在骨组织中的定位,用Westernblot杂交法检测骨组织TGFβ1、Smad23蛋白表达。结果与对照组(Sham组)比较,模型大鼠(OVX组)骨组织Tb.Th缩小,%Ma.Ar明显增大,差异有显著性(P<0.01);OVX组TGFβ1的mRNA、蛋白及Smad23蛋白表达水平显著降低(P<0.01),Smad23蛋白广泛表达于骨组织干骺端,主要表达于骨基质表面及骨小梁周围的成骨细胞。结论TGFβ1及其信号转导蛋白Smad23可能在绝经后骨质疏松症中起重要调节作用。
Objective To observe the expression and significance of transforming growth factor β1 (TGF-β1) and its signal transduetion protein Smad 2/3 in the bone tissues of OVX rats. Methods The model of primary osteoporotic rat were established by ovariectomy. After 13 weeks of the operation, The right tibia was removed and bone histomorphometry, including trabecular area, trabecular thickness and number trabeculae, were measured ; RT-PCR was applied to test the the expression of TGF-β1 mRNA in the bone tissues and immunohistochemistry and Western blot for Smad 2/3 protein. Results The bone histomorphometric data decreased significant in OVX group ( P 〈 0.01) ;The expression of TGF-β1 mRNA and its protein and Smad 2/3 protein reduced greatly (P 〈 0.01 ). The Smad 2/3 protein was widely expressed at the epiphysis of the bone tissue, and strongly expressed in the osteoblasts on the surface of the bone matrix and around the bone trabeculae. Conclusions The descending regulation of the expression level of TGF-β1 mRNA and its signal transduction protein Smad 2/3 may be an important mechanism for priraary osteoporosis.
