摘要
目的研究体外培养的骨膜细胞的形态学特征。方法采用常规细胞培养法进行骨膜细胞体外培养,并观察其形态特征及超微结构。结果人骨膜培养活细胞光镜下形态:早期新生细胞呈梭形,饱满透明,立体感强;分裂期呈立方形或短柱状;后期由长梭形逐渐变成宽梭和不规则形,分泌功能旺盛。人骨膜细胞电镜超微结构:胞体呈梭形,膜面有少量微绒毛状突起,粗面内质网丰富,有大量异噬体等次级溶酶体,线粒体体积较小、基质致密有少量嵴。具有原代细胞生物形态学特征。结论人骨膜成骨细胞可经体外培养大量增殖,且细胞传代进化后,其形态学结构与原代细胞无明显差异,可用于细胞移植。
Objective To study the morphological characteristic of human periosteal cells cultivated in vitro. Methods To observe the morphological characteristic of human pefiosteal cells cultivated by the conventional method in vitro under optics and electronic microscope. Results Under optics microscope: neonatal periosteal cells showed shuttle, satiation, clarity and third dimension in early stage, then become cube or puncheon in split stage, and gradually become wide shuttle and irregular shape with a lot of secretion in telophase. Under ultrastructure, the bodies of these cells show fusiform, and there are a lot of microvilli on membranous wall and abundant endoplasmic reticula. The volume of mitochondria whose matrix has a few cristallin is less than that of normal pcriosteal cells. Conclusion Human pcriosteal cell can be greatly cultivated and proliferate in vitro. Those cellular configurations have no distinct difference with regeneration cells after anagenesis, the cultured cells can be used in transplantation.
