摘要
目的:探讨急性癫痫大鼠模型脑海马中碱性成纤维细胞生长因子(bFGF)的表达、分布及其意义。方法:将39只健康雄性Wistar大鼠随机分为正常对照组(A组)3只、生理盐水组(B组)和癫痫模型组(C组)各18只,A组不处置,C组大鼠以氯化锂-匹罗卡品诱导急性癫痫模型,B组以相同体积的生理盐水代替匹罗卡品,A组大鼠处死,B、C组分别于造模后6、12、24 h及3、71、4 d时各处死3只,检测各组大鼠海马中bFGF的表达规律及其在海马细胞中的分布情况。结果:造模后A和B组海马区发现有bFGF蛋白的表达,但表达水平较C组低(P<0.05);C组癫痫发作后6 h海马区bFGF表达开始增加,24 h达高峰,以后逐渐下降,14 d时仍维持较高水平(P<0.05);bFGF主要表达于星形胶质细胞,CA2区部分神经元表达。结论:bFGF在急性癫痫大鼠模型脑海马中呈规律性表达,以星形胶质细胞表达为主,CA2区部分神经元表达;bFGF可能对癫痫后的脑损伤起保护作用。
Objective:To study the expression of basic fibroblast growth factor (bFGF) in the hippocampus of epileptic rat at different time points and to investigate the relationship between bFGF and epilepsy. Methods: Thirty-nine adult male Wistar rats were randomly divided into three groups: normal control group, NS group and epilepsy model group. Lithium-pilocarpine induced seizure models were used and the expression and distribution of bFGF in the hippocampus of rats were studied by immunofluorescent labeling techniques. Results: bFGF was found in control group and NS group, but the expression was lower than in epilepsy model group(P^0.05). In the model group, the expression of bFGF began to increase 6 h after epilepsy onset, reached a peak after 24 h, then declined, but it still kept at a high level after 14 days (P〈0.05). Double immunofluorescent labeling revealed that, in the hippocampus, bFGF immunoreactivity was mainly expressed in astrocytes, but some neurons in CA2 area also showed bFGF immunoreactivity. Conclusion: bFGF was expressed regularly in the hippocampus of acute epileptic rats and was mainly detected in astrocytes, bFGF may protect the brain injury following seizure in rats.
