Significance of nm23 and E-cadherin protein expression in colorectal carcinoma

摘要 目的探讨nm23和E-cadherin在大肠癌转移进展过程中的作用及相互关系。方法用免疫组化EnVision二步法分别检测186例大肠癌、癌旁组织和20例正常大肠粘膜nm23和E-cadherin蛋白的表达情况,结合临床病理资料进行分析。结果nm23和E-cadherin蛋白在大肠癌组织的阳性表达率分别为52.7%、56.5%,显著低于癌旁组织(分别为90.99%、95.2%)和正常大肠粘膜(分别为95%、100%);nm23和E-cadherin在高分化腺癌的阳性率高于中-低分化腺癌;随着Dukes分期从A期到D期,淋巴结的转移,nm23和E-cadherin的表达显著降低或缺失;nm23和E-cadherin在大肠癌的表达呈正相关。结论nm23和E-cadherin表达的降低或缺失与大肠癌的恶性进展,转移具有密切关系,检测大肠癌组织中的nm23和E-cadherin的表达水平对预测患者的预后具有重要意义。 Objective To investigate correlations of expressions of nm23 and E - cadherin in coloreetal carcinoma to metastasis and progress, and their interrelations. Methods nm23 and E - eadherin expressions were detected by immunohistochemieal method (Envision two - step method) in paratfin- embedded sections from 186 eases of colorectal carcinoma and adjacent non - eaneeros tissues, 20 eases of normal eolorectal tissues, the relationship between their expressions and clinieopathologie data were analyzed. Results The positive rate of nm23 and E- eadherin in coloreetal carcinoma were 52.7% and 56.5% respectively, which were significantly weaker than those in adjacent non - cancerous tissues and normal tissues. The expressions of nm23 and E - eadhefin were significantly lower in metastatic group ( P 〈 0.01 ). Positive correlation existed between the expression of nm23 and E - cadhefin. Conclusion Down - regulations of nm23 and E - cadherin closely relate to metastasis of eolorectal carcinoma. Detection of nm23 and E - cadherin might be helpful to predict prognosis of coloreetal carcinoma patients.
