摘要
采用随机扩增多态性DNA(RAPD)技术,利用17个引物对26个安徽野生香菇菌株和6个香菇栽培品种进行了遗传多样性分析,并构建了遗传相关聚类图。在128个RAPD标记中,多态性标记为104个,占81.3%。聚类分析显示,当以相异距离0.25为阈值,32个菌株被划为5个RAPD遗传组,多数菌株之间遗传相似性较低。表明供试菌株在DNA水平上存在比较显著的遗传变异,具有较丰富的遗传多样性。杂种优势的检测表明亲本间遗传距离较大的组合其杂种优势也较强。
Using technique of random amplified polymorphic DNA (RAPD) assay 26 wUd stratus trom Anhui province and 6 cultivated strains of Lentinula edodes in China were analyzed for genetic diversity with 17 primers. Among 128 amplified fragments 104 (81.3%) were polymorphic. In 0.25 dissimilarity distance cluster analysis revealed the existence of five distinct genetic groups in which group. The results show that genetic similarity of these strains was low and there were obvious genetic variations in DNA level. The resuits revealed that high productive hybrids were obtained by crossing parents with larger genetic distances.
