AF—DX116对犬的呼吸兴奋效应

摘要 犬静脉注射AF-DX1160.05,0.5,5mg/kg均导致①呼吸频率、潮气量、每分通气量明显增加(P<0.05或0.01),均呈剂量依赖关系。②PaO2升高,PaCO2降低。0.20mg/kg椎动脉注射亦产生与静脉给药相似的呼吸兴奋效应。AF-DX116还能拮抗6β-AN的呼吸抑制作用,表明,AF-DX116有呼吸兴奋作用,并可能与其阻滞呼吸中枢M2受体有关。 The excitatory effect of AF-DX 116 on respiration and its relation to M2-receptors Were studied in dogs.It was found that the respiratory frequency(FR),ventilation volum.(Vm)and PaO2 were increased and the PaCO2 was decreased following administration of AF-DX 116 0.05~5mg/kg iv. The excitatory effects on FR and Vm were dose-dependent in manner.when small doses of AF-DX 116 were given by ia route,an excitatory effect was observed similar to the result following the drug given by iv route. AF-DX 116 given either by iv or ia route, antagonized the inhibitory effect of 6β-AN, which stimulates M2 receptors,It appears that the excitatory action of AF-DX 116 on respiration might be related to the block of inhibitory M2 cholinergic receptors in the respiratory center.
