人促甲状腺激素单克隆抗体杂交瘤细胞株的建立 被引量:2

摘要 以进口纯人促甲状腺激素(h—TSH)作免疫原,经用Galfre等杂交瘤制备法,分别制备成13B4、13C4、8H10、3E4四株具有抗h—TSH特异性,与促黄体素、促卵泡素、绒毛膜促性腺素无明显交叉反应。具有稳定分泌功能的杂交细胞株。经亚型鉴定均为IgG1。液相中与125I—hTSH的最大结合率依次为13B4、3E4、8H10和13C4,分别为61%、59%、56%和34%。表位分析表明,4株单抗主要针对两个不同表位。在此基础上已成功地建立h—TSH固相免放和酶免吸附超敏测定法。 Four hybridoma cell lines-13B4,3E4,8H10 and 13C4 are established while purified human thyrotropin is used as immunogen,The monoclonal antibodies produced are highly specific for two separately located epitopes of h-TSH and are all IgG1. Little or no cross-reaction with h-LH,h-FSH and hCG is observed. Binding capacities of the antibodies with125I-hTSH in liquid phase are 61%(13B4),59%(3E4),56%(8H10) and 34%(13C4). With those antibodies,uItra-sensitive IRMA and ELISA for h-TSH have been established.
