摘要
目的:观察黄连解毒汤有效部位(HLJDTAF)对培养皮层神经细胞过氧化氢损伤的保护作用。方法:体外培养新生大鼠皮层神经细胞,加入过氧化氢观察其对神经细胞的损伤及HLJDTAF的保护作用。结果:HLJDTAF 610,305 mg·kg-1可减少模型乳酸脱氢酶(LDH)漏出率,305 mg·kg-1还可提高模型细胞活性(P<0.05;P<0.01)。同时HLJDTAF各剂量均可减少丙二醛生成,153 mg·kg-1还可减少一氧化氮生成。结论:HLJDTAF对培养神经细胞自由基损伤具有显著的保护作用,其机制可能与抗脂质过氧化作用有关。
OBJECTIVE To investigate the protective effects of Huanglian Jiedu tang active fraction(HLJDTAF) on hydroperoxide(H2O2 )-induced injury in cerebral cortical neuronal cultures. METHODS Cortical neurons of newly born rat were cultured in vivo. The protective effects of HLJDTAF on hydroperoxide-induced injury were observed. RESULTS HLJDTAF 610,305 mg·kg^-1 reduced the leakage rate of LDH, 305 mg·kg^-1 increased the activity of neuron, 610, 305, 153 mg·kg^-1 reduced the production of MDA. CONCLUSION HLJDTAF protects hydroperoxide-induced injury in cerebral cortical neuronal cultures , the mechanism is probably related to the suppression of the generation of NO and reduction of lipid peroxide action.
