硒与氟对体外培养的人头皮毛囊丙二醛水平的影响

摘要 目的观察离体培养条件下氟与硒对人头皮毛囊脂质过氧化物水平的影响。方法构建人头皮游离毛囊培养模型,在模型中,加入不同浓度的氟化钠和亚硒酸钠,筛选浓度后分为7组;用硫代巴比妥酸(th ibab ituric ac id,TBA)法测定毛囊培养基中丙二醛(MDA)含量,并进行统计分析。结果0.1 mmol/L氟化钠不能增加游离毛囊培养基中MDA含量,1 mmol/L和10 mmol/L的氟化钠能增加MDA含量;0.01 mmol/L的亚硒酸钠能拮抗1 mmol/L氟化钠的上述作用,但不能拮抗10 mmol/L氟化钠对MDA水平的影响。结论一定浓度的氟化钠能增加培养基中MDA的水平,0.01mmol/L亚硒酸钠能拮抗一定浓度的氟化钠对毛囊培养基MDA水平的影响。 Objective To observe the effect of fluorine and selenium on the lipid peroxide(LPO) level of human hair follicle in vitro. Methods The single hair follicle from human scalp was separated and cultured, then added in different concentrations of sodium fluoride and sodium selenite. Chosen the appropriate concentrations, they were divided into 7 groups. The TBA method was used to determine the concentrations of MDA in culture medium. Results 0.1 mmol/L sodium fluoride did not increase the content of MDA, but 1 mmol./L and 10mmol/L sodium fluoride increased the content of MDA. 0.01 mmol/L sodium selenite weaken the effect of 1 mmol./L sodium fluoride on MDA level,but did not weaken the effect of 10 mmol/L sodium fluoride. Conclusion Sodium fluoride of certain concentration increased the MDA level of hair follicle's culture medium, 0.01mmol/L sodium selenite weakened the effect of sodium fluoride of certain concentration.
