家兔上矢状窦中1/3及其回流静脉结扎后脑水肿变化和水通道蛋白-4表达的研究 被引量:5

摘要 目的探讨家兔上矢状窦中1/3及其回流静脉结扎后窦旁皮层组织水肿变化、水通道蛋白-4(AQP4) 的表达及其变化。方法建立家兔上矢状窦中1/3及其回流静脉结扎的动物模型,用干湿重法、免疫组化方法和半定量逆转录聚合酶链反应(RT-PCR)分别检测窦旁皮层组织脑水肿、AQP4蛋白和AQP4mRNA的表达及其变化。结果家兔上矢状窦中1/3及其回流静脉结扎后,家兔可长期存活而无死亡;窦旁皮层组织4 h出现脑水肿,8 h到达高峰,以后随时间延长而逐渐减轻,长期组脑水肿接近于假手术对照组。家兔上矢状窦中1/3及其回流静脉结扎后窦旁皮层组织存在AQP4蛋白的表达,结扎后4 h AQP4表达显著高于正常水平,8 h达到峰值,其表达水平以后随时间延长而逐渐回落。AQP4的表达与脑含水量呈显著正相关(r=0.778,P<0.01);而窦旁皮层组织也存在 AQP4mRNA的表达,其结果与AQP4蛋白的表达相似。AQP4mRNA的表达与脑含水量呈显著正相关(r=0.885,P <0.01)。AQP4mRNA与AQP4蛋白的表达呈显著正相关(r=0.753,P<0.01)。结论家兔上矢状窦中1/3及其回流静脉结扎后窦旁皮层组织存在脑水肿,AQP4蛋白与AQP4mRNA在兔脑中有表达。两者的表达增强可能与上矢状窦中1/3及其回流静脉结扎后脑水肿的形成和发展密切相关。AQP4可能参与结扎后脑水肿的形成。 Objective To investigate the brain edema and the expression of aquaporin -4 of the cerebral cortex in the middle third of the superior sagittal sinus and the reflux veins occlusion in rabbits. Methods The model of the middle third of the superior sagittal sinus and the reflux veins occlusion in rabbits was made. Dry-wet method was used to determine the brain edema. Immunohistoehemistry was used to determine the change of expression of AQP4 protein. The expression of AQP4mRNA was assessed by RT-PCR. Results The rabbits can live for a long period after the middle third of the superior sagittal sinus and the reflux veins occlusion. The brain edema of the cerebral cortex increased after 4 hours, and increased significantly after 8 hours, then decreased gradually with time prolonged, the brain edema of the long period group was similar to the shamed operation group. There was AQP4 protein expression in rabbits brain, increased after 4 hours, get the highest level after 8 hours ,and then decreased gradually with time prolonged. The AQP4 protein expression was positive linear correlated with the brain water content( r =0. 778, P 〈0.01 ). There was AQP4mRNA expression in rabbits brain too, the result was the same as the AQP4 protein expression . The expression of AQP4mRNA was positive linear correlated with the brain water content(r=0.885, P〈0.01) and the AQP4 protein(r =0.753, P〈0.01). Conclusions There is brain edema after the middle third of the superior sagittal sinus and the reflux veins occlusion . There are AQP4 protein expression and AQP4mRNA expression in rabbits brain after the operation. The increased expression of them perhaps is closze related with the brain edema formation and development after the middle third of the superior sagittal sinus and the reflux veins occlusion. AQP4 may participate in the brain edema formation .
