摘要
目的:建立以酸性染料比色法测定苦参碱脂质体中苦参碱含量的方法.方法:将苦参碱脂质体上样于用蒸馏水溶胀 12h以上的葡聚糖凝胶 G-50柱,以蒸馏水为流动相洗脱后与 0.0 125%溴代麝香草酚蓝缓冲液、氯仿混合振摇,静置分层,并于 413nm波长处测定氯仿层吸收度.结果:苦参碱检测浓度在 0.04~ 0.20mg/ml范围内与吸收度线性关系良好(r=0.9 972),平均加样回收率为 94.10%(RSD=1.86%).结论:本方法操作简便、准确,可用于苦参碱脂质体的质量控制.
OBJECTIVE:To establish a method for content determination of matrine in it's liposomes by acid dye colorimetry. METHODS: Sephadex gel G- 50 column was established by swelling of matrine liposomes sample in distilled water for at least 12 hours, distilled water was used as mobile phase, after eluting, the column was mixed with 0.0 125% of bromothymol blue buffer solution and chloroform,and then the mixture was shaken, standing and demixed, the absorbability of chloroform layer was detected at the wavelength of 413 nm.RESULTS: The detection concentration of matrine showed good linearity with its absorbability within the range of 0.04-0.20mg/ml( r=0.9 972), the average recovery was 94.10% (RSD : 1.86% ). CONCLUSION:The established method is simple and accurate,which can be used for quality control of matrine liposomes.
