摘要
用异型双功能连接剂SPDP,将核糖体抑制蛋白一天花粉蛋白(TCS)共价连接在单克隆抗体ND-1上,制成免疫毒素,此单抗与人大肠癌细胞表面抗原一大分子表面抗原(LEA)有高度亲合能力。连接物ND-1-TCS用SephadexG-150分子筛层析纯化。间接免疫荧光试验结果表明,ND-1-TCS保留了较高的抗体活性。体外细胞毒性试验表明,ND-1-TCS对人大肠癌细胞CX-1的IC_(50)。为0.36μg/ml.细胞毒性ND-1和TCS混合物高10倍。ND-1-TCS对人大肠癌细胞的细胞毒性比非抗原携带细胞SP_(2/0)高60倍。此免疫毒素在人大肠癌的治疗方面有应用前景。
ribosome inactivating protein-trichosanthin(TCS ) was covalently linked to a monoclonal anti-body ND-1to prepare a kind of immunotoxin by heterobifunctional reagent SPDP,which exhibited highaffinity for cell surface antigen large external antigen (LEA)of hurnan gut carcinoma cells. The conju-gant ND-1-TCS(Immunotoxins were purified on Sephadex G-150 column, The results of indirect im-munofluo-rescent test showed that ND-1-TCS was retained very high antibody activity. The in vitro cy-totoxicity assays showed ND-1-TCS killed human gut carcinoma CX-1 by 50%(IC_50)at concentrationof 0.36μg/ml.The cytotoxicity was 10-fold higher than that of the mixture of free TCS and ND-1.The cytotoxicity of ND-1-TCS to CX-1 cell was 60-fold higher than that of ND-1-TCS and TCS toLEA-negative carcinoma cell SP_2/0.The result indicated that the immunotoxins may be proved useful inthe treatment of human gut carcinoma.
