莫达非尼对睡眠剥夺汽车兵汽车驾驶能力和疲劳感的影响 被引量:8

Effects of modafinil on driving performance and fatigue level of military truck drivers during 36h sleep deprivation

摘要 目的观察36 h睡眠剥夺(SD)条件下服用莫达非尼对汽车兵汽车驾驶能力和疲劳感的影响。方法60名男性汽车兵随机分为安慰剂组和莫达非尼组(各组n=30)。试验第1日起床(6∶00)后,36 h保持不睡眠,于当日夜间23∶00、第2日5∶00、11∶00三次服用莫达非尼或安慰剂200 mg。于试验第1日8∶30和14∶30及用药后4个时刻(即试验第2日7∶00、10∶00、13∶00、16∶00)共完成6次驾驶任务。测定指标包括考核场汽车驾驶能力、视听反应能力和临界闪光融合频率(CFF)、斯坦福嗜睡量表和自认疲劳分级量表评分。结果两组的汽车驾驶成绩和视听反应能力无明显差异。与安慰剂组相比,莫达非尼组的CFF明显增加、主观嗜睡和疲劳感显著降低。结论36 h SD条件下分次服用常规剂量的莫达非尼具有显著降低机体疲劳感的作用,且对汽车驾驶能力无明显不良影响。 Objective To investigate the effects of modafinil on driving performance and fatigue level of military truck drivers during 36 h sleep deprivation. Methods Sixty male military truck drivers were randomly divided into placebo and modafinil groups (n=30). After getting up at 6 : 00 in the first day, they were required to finish the experiment for 36 h without sleeping, and modafinil (200 mg) or placebo was given at 23 : 00 of the first day and 5 : 00 and 11 : 00 of the second day. Six driving tasks were tested on testing ground at 8 : 30 and 14 : 30 of the first day and 7 : 00, 10 : 00, 13 : 00, 16 : 00 of the second day respectively. The following parameters were measured, driving performance on the testing ground, visual and auditory reaction time, critical flicker frequency(CFF), subjective scores of Stanford sleepiness scale(SSS) and rating of perceived exertion(RPE). Results The differences of driving performance and visual and auditory reaction time between two groups were non-significant. Compared with placebo group, CFF in modafinil group was significantly increased and subjective sleepiness and fatigue level were remarkably decreased. Conclusions Repeated oral administration of modafinil at routine dose could significantly decrease the subjective fatigue level during 36 h sleep deprivation, and had no adverse effects on driving performance.
